Transcription is an essential and highly regulated cellular process that is carried out by RNA polymerases. There are three types of RNA polymerases in Saccharomyces cerevisiae, each specialized in the synthesis of different transcripts. Apart from that, prefoldin-like Bud27 is a cochaperone involved in multiple cellular functions. In the present work, we demonstrate the role of Bud27 as a coordinator of the activity of the three RNA polymerases for ribosome biogenesis. In addition, the function of Bud27 during the regulation of transcriptional repression of RNA pol III, responsible for the synthesis of transfer RNA and 5S ribosomal RNA, has been characterized. This regulation is exerted by Bud27 through the control of PP4 phosphatase activity and the repressor Maf1. Finally, the role of Bud27 on the composition of the RNA pol III transcriptional machinery was further explored.
